Differentiation to Definitive Endoderm

Human induced pluripotent stem cells (hiPSCs) were grown on Vitronectin XF™ plates. After 5 passages the cells were replated on Vitronectin XF™ and subjected to Definitive Endoderm (DE) differentiation using a protocol adapted from Si-Tayeb et al, 2010, Hepatology 51:297. After the initial 5 day exposure to Activin A/BMP-4/FGFb, the cells were harvested and analyzed by flow cytometry for the DE markers GATA4, CXCR4, and FoxA2, and for the absence of the pluripotency marker Oct4.

Hepatocyte Differentiation

Human pluripotent stem cells (hESCs and hiPSCs) were plated on Vitronectin XF™ and subjected to hepatocyte differentiation using a protocol adapted from Si-Tayeb et al, 2010, Hepatology 51:297. After the initial 5 day exposure to Activin A/BMP-4/FGFb, the cells were stained for presence of GATA4 and absence of Oct4. Data courtesy of M. Nagaoka and S. Duncan, Medical College of Wisconsin.

Cells were fixed and stained at Days 10, 15, and 20 of the hepatocyte differentiation protocol. Cells expressing hepatocyte markers HNF4A (red), alpha-fetoprotein (AFP; green), and Albumin (ALB; green) were detected by immunocytochemistry. Nuclei were counterstained with DAPI.
